Abstract: the present disclosure relates to bifunctional compounds comprising targeting moiety that binds swi/snf related, matrix associated, actin dependent regulator of chromatin, subfamily a, member 4 (smarca4) or smarca2 and smarca4, and a degron that binds an e3 ubiquitin ligase. the disclosure further provides compositions and methods for treating diseases or conditions mediated by aberrant activity of smarca4 or smarca2 and smarca4, including diffuse midline glioma harboring a h3k27m mutation.
