Abstract: described herein are devices and methods for fluid manipulation using electrowetting. more specifically, this disclosure provides devices and methods for manipulating fluids using a digital microfluidics (“dmf”) device. for example, a fluid may be provided to a reservoir of a dmf device. the reservoir of the dmf device may include an electrode array configured to discretize the fluid into more than one large-volume droplets, each of which may be individually manipulated within the reservoir. any one of the more than one large-volume droplets may be primed or split into one or more medium-volume droplets. the primed droplets or medium-volume droplets may further be split into one or more small-volume droplets for further processing on the dmf device. accordingly, the described devices and methods enable high throughput and space savings on the dmf device.
