this invention relates to processes and systems for flow-through solid-phase oligonucleotide synthesis. the process comprises the steps of detritylation. coupling. oxidation, and capping. the process comprises at least one step of reconsolidating the synthetic resin that occurs after the detritylation step. the step of reconsolidating the synthetic resin comprises a first fluidizing step followed by a repacking step. in the fluidizing step solvent flow from the bottom of the column to the top, and in the repacking step solvent flow from the top of the column to the bottom.
